Yes, the testdef file is required, no question. In the test
definitions, there, you reference the "requester" modules.
Therefore, there are tests that do not need their own requester,
since they e.g. use SNMP or whatever else already existing
requester.

This is definitely not needed. In case of realhttp.pm there is a
Monitor module because there are two implementations in parallel
- the old one using Monitor::realhttp and the new one using testdef
and Requester.

Not needed, either. uxmon/Config/* and bin/Monitor/* are used for
old-style test implementations, only.

This is needed if (and only if) the testdef configuration references
a requester called "temperature", e.g. by doing something like
get temperature.currentTemp;
somewhere (=> get system parameter currentTemp via the "temperature"
requester).
realhttp achieves this by
...
set domain realhttp;
...
set var ${domain};
get ${var}.status.${id} ...
...

This is required if you want the server (bsmon) to create graphs for
your test.

No, that's all.
Better than searching an installed Big Sister have a look at the
"modules" directory in the source code. The realhttp sub directory,
for instance, contains everything, that is required for the new-style
realhttp test.
